2 транскрипционно-опосредованная амплификационная система
[лат. transcriptio — переписывание; лат. amplificatio — распространение, увеличение; греч. systema — целое, составленное из частей, соединение]изотермическая реакция амплификации нуклеиновых кислот, обладающая более высокой чувствительностью по сравнению с полимеразной цепной реакцией (см. полимеразная цепная реакция). Каждый цикл этой реакции состоит из двух этапов: а) синтез двухнитевой кДНК, при котором в состав вновь синтезированных молекул вводится нуклеотидная последовательность, распознаваемая ДНК-зависимой РНК-полимеразой (чаще всего Т7-полимеразой) и б) амплификация целевой нуклеотидной последовательности с помощью РНКполимеразы. При использовании для амплификации ДНК или РНК Т.о.а.с. в течение 40—50 мин образуется 10 8 — 10 9 целевых копий. Первое сообщения об этом методе было сделано Д.
